Instructions for Creating a Clone Request File

The clone selection file downloaded from the PFGRC website is in tab–delimited format with a ".txt" extension. Tab-delimited format separates all columns by tabs. All clone request files uploaded to the website should also be in tab–delimited format.

The first row in the clone selection file should contain a heading. The heading line must start with the # character.

The first column of the clone selection file should contain locus information from the #TIGR Locus# column of the file you downloaded from the PFGRC on the Clone Selection page. Additional information can be present in columns after the first column, however, we ignore data in any other columns.

A separate file should be made for every organism which you want to order clones from.

Summary of requirements for the clone request file:

  • Tab-delimited format
  • Contains header line that begins with #
  • First column contains loci from the #TIGR Locus# column
  • Each locus on a separate row
  • Loci come from a single organism

The following illustrates how to create a clone request file from the clone selection file downloaded from the PFGRC using Microsoft Excel.

Making a clone request file with Microsoft Excel

  • To open the clone selection file using Microsoft Excel, either right–click on the .txt file and the select "Open with–>Microsoft Excel" or open Microsoft Excel and select "File–>Open" then choose the .txt file.
  • To create a clone request file open a new file in Microsoft Excel.
  • For each clone being requested, add the ORF locus# to column A and a single row. This can easily be done by copying and pasting the ORF locus# from the clone selection file to the clone request file.
  • To save the clone request file select File–>Save As and enter a file name. Under the 2nd dropdown box entitled "Save as type:" select "Text (tab–delimited) (*.txt)" and then "Save".
